I am planning to build a class E SSTC.I had read Steve's page already.
But I want to know if there is any way to calculate the value of tank capacitor C6?Can I start with a theoretical value and fine tune it instead of totally try and error?
I have a oscilloscope but I only has one probe.Can I tune it with only one probe? Or I should buy another probe?

It's easier to test it since there are so many stray capacitances and inductances guaranteed to show up in your setup.
Remember though that to calculate it you need to first find out what inductance shows up in the tank - that's another calc to make first. Might as well just measure things from the get-go.
Also remember that MOSFETs have a significant drain-source capacitance (which does vary)
You want your capacitor in parallel to be somewhat larger then this D-S capacitance so that you have a little more stability in your tank (the parallel fixed capacitor has more effect on the resonant frequency then the somewhat variable DS capacitance, it overwhelms it slightly).
