You can vanish it before winding if you want to. I found that it is not necessary if it is pvc. If it was cardboard, i definately would varnish before winding. Just make sure everything is clean.

I usually sand off the outer shiny layer of the PVC, then clean and varnish it. Honestly varnishing before winding is probably necessary, no performance difference (that I have seen). However one "bonus" of varnishing first is that it will cause the wire to stick to the coil form if you don't let it dry completely, can save you if you accidentally loose wire tension while winding.
